EXPERIMENTAL DESIGN

WHAT IS OPTIMAL AND WITH WHAT IS POSSIBLE 


Based on your material and scientific question, together we discuss different options for the experimental design, e.g. the appropriate number of samples/replicates, optimal storage and handling, and choice of sequencing technique. 

PRECIOUS SAMPLES


We specialize in sequencing of difficult samples. Often samples with low RNA content are the most precious, so why not let us unfold the potential of your precious samples by performing miRNA or total RNA sequencing? Low input samples include plasma, exosome purification, cerebrospinal fluid (CSF), laser microdissected tissue and more.



AVOID BIASES


It is important to consider what sources of bias your experiment could have. These could be sex, age, the time since samples were taken, sample quality and much more.

Groups to be compared by NGS must be as similar as possible in the potential bias-causing attributes to ensure that significant hits are real and not caused by bias.




SEQUENCING

After an initial discussion, we will make a plan for library preparation and sequencing. In general, the amount of your sample and quality of the RNA are central factors for which preparation method is the optimal choice. The final libraries produced are quality controlled before moving on to sequencing.

TOTAL RNA


Open up the biology of your samples by exploring the transcriptome in great detail.


Our best selling product is the complete quantification of long RNAs in customer samples. This includes mRNA and long noncoding RNA species. Our standard total RNA-seq package includes differential expression analysis and publication grade plotting of results. We also specialize in analysis of circular (circRNA) from total RNA data.

SMALL RNA 


Precise quantification of small RNA



Small RNAs provide vital cellular control mechanisms. In disease states and during developmental processes, the network of miRNAs and other small RNAs is often affected. Gaining insight into the miRNA expression signature in cells/tissue/fluid of interest can have great impact on biomarker discovery and insight into mechanisms involved and much more.


Small RNA sequencing can be used for analysis of: miRNA, tRNA, siRNA, piRNA, snoRNA, vaultRNA and more

BIOINFORMATICS

At the initial consultation, we also discuss what bioinformatic analysis would be relevant for your question. In the case of standard analysis, we have a defined package of figures and tables to give you both easy overview and the catalogue to zoom in on details on single transcripts.
